There's a woman at the office that I work in that I have a strong interest in. When ever we meet, we usually stop and chat a little while. Each time we do so, she will slip her foot out of her shoe for a few seconds, and then slip it back in. Is this a signal from her, or do her shoes just hurt her feet?

This falls into the category of "Only She Knows". There is no way that anybody else in the world could possibly know!! Maybe this is a casual thing she does when she's relaxed, to rest her feet. Maybe her feet kill her and she feels comfortable around you enough that she can finally give them a break.

You need to focus more on the talks you guys have, the sharing of stories, and maybe start having drinks or dinners together to build up your friendship. You can't really determine anything about a person by their taking off of shoes :)
